Anti-GPA33 immunotoxin (scFv)-α-sar (CAT#: AGTO-G034F)
This immunotoxin was achieved by conjugating/fusing the Anti-GPA33 scFv to α-sarcin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to GPA33 tumor antigen expressing cells. Once bind, alpha-Sarcin enters into the cells by receptor-mediated endocytosis and interacts with a single bond in the targeted ribosome and breaks it, causing the ribosome to be inactivated. It was designed for treatment of colon cancer.

- Alpha-Sarcin is a ribosome-inactivating protein produced in the mold Aspergillus giganteus. Specifically, alpha-sarcin cleaves the a portion of ribosomal RNA that forms the small ribosomal substrate. The high specificity of alpha-sarcin and its efficiency of cleavage are point of study and also account for this protein's very high toxicity level.
