LAM is an important immunodiagnostic target for detecting active infection with Tuberculosis (TB). This recombinant mouse antibody Fab fragment (CS-35) is capable of binding to LAM. CS-35 may interact with additional Araf residues connected through α-(1→5)-glycosidic linkages to the reducing end of residue A. Such residues are found in natural LAM and may enhance binding affinity to CS-35.

SDS-PAGE analysis of PFBW-158 in β-mercaptoethanol-reduced (Lane 2) and non-reduced (Lane 1) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result of different β-mercaptoethanol-reduced proteins (Heavy chain and Light chain) migrate as about 27 kDa and 25 kDa, respectively.

Western blot analysis of PFBW-158 was performed by loading Native M. Tuberculosis LAM-Antigen.
PFBW-158 incubation concentration: 2 ng/μL.
The secondary antibody: HRP-conjugated Goat Anti-Mouse IgG ( H+L).
Lane 1: Reducing Antigen (0.5 μg)
Lane 2: Reducing Antigen (1 μg)
Lane 3: Reducing Antigen (2 μg)

ELISA analysis of PFBW-158 was performed by coating with Native M. Tuberculosis LAM-Antigen. Then blocked with BSA and incubated with PFBW-158. The HRP-conjugated Mouse Anti-His as a secondary antibody (1: 2000). Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.
