This product is a recombinant chimeric antibody that recognizes Siglec-15. Siglec-15 was found to be expressed in several cancer types, in particular ovarian cancer, renal cancer, cancer of the central nervous system, and prostate cancer. Recombinant human Siglec-15 was used as an immunogen for the mAb generation.

SDS-PAGE analysis of TAB-445CT in non-reduced (Lane 1) and reduced (Lane 2) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result of different β-mercaptoethanol-reduced proteins (Heavy chain and Light chain) migrate as about 50 kDa and 25 kDa, respectively. And non-reduced protein migrates as 180 kDa.

Western blot analysis of TAB-445CT was performed by loading Human Siglec-15 / CD33L3 Protein (His Tag) onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least one hour. Membranes were probed with TAB-445CT and HRP Goat Anti-Human IgG as a secondary antibody. Chemiluminescent detection was performed.

Dot Blot analysis of TAB-445CT was performed by coating with Human Siglec-15 / CD33L3 Protein (His Tag).

TAB-445CT incubation concentration: 2 μg/mL.
HRP-conjugated goat anti-Human IgG as a secondary antibody (1: 2000)

ELISA analysis of TAB-445CT was performed by coating with Human Siglec-15 / CD33L3 Protein (His Tag). Then blocked with BSA, and incubated with TAB-445CT. The HRP-conjugated goat anti-human IgG as a secondary antibody. Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.

Effective In Flow Cytometry

We used this chimeric antibody to detect Siglec-15 expression on the surface of tumor cells. The flow cytometry results were clear and reproducible. It is a great reagent for studying sialic acid-binding Ig-like lectins in cancer research models.

Useful Blocking Antibody

This antibody served as an excellent blocking agent in our functional assays. It allowed us to investigate the role of Siglec-15 in immune regulation. The quality is high, with SDS-PAGE confirming purity over 95 percent.
