The anti-ADORA2A pH-dependent antibody (clone 864H1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human ADORA2A. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for ADORA2A under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from ADORA2A at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. ADORA2A (Adenosine A2A Receptor) is a G protein-coupled receptor that acts as an important immune checkpoint molecule, regulating T-cell function in the tumor microenvironment. It is also a key regulator of brain function, playing a role in motor control and mood.
