The anti-CEACAM1 pH-dependent antibody (clone 18-20) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CEACAM1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CEACAM1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CEACAM1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CEACAM1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CEACAM1 is a cell adhesion molecule and an inhibitory receptor expressed on various immune and epithelial cells. It plays a role in regulating the immune response and is also involved in cancer development and progression.
