The anti-CXCL8 pH-dependent antibody (VS-0825-YC93) targets both human and monkey CXCL8 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CXCL8 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CXCL8 clearance. The antibody operates in a cyclical manner, binding CXCL8 in the extracellular fluid and releasing it in acidified endosomes. CXCL8, also known as Interleukin-8 (IL-8), is a potent chemokine that attracts and activates neutrophils. It is a central mediator of inflammation and is implicated in a wide range of inflammatory diseases.
