The anti-DDR2 pH-dependent antibody (VS-0825-YC98) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human DDR2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for DDR2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from DDR2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for DDR2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human DDR2, or Discoidin Domain Receptor 2, is a receptor tyrosine kinase that binds to collagen. It is involved in cell-matrix interactions and is a key player in the development of fibrotic diseases and in the progression of certain cancers.
