The anti-EGFRvIII pH-dependent antibody (VS-0825-YC111) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human EGFRvIII. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for EGFRvIII under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from EGFRvIII at pH 6.0. This pH-sensitive antibody retains its binding affinity for EGFRvIII on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human EGFRvIII is a constitutively active mutant of the EGFR protein that lacks a portion of its extracellular domain. It is a common mutation in glioblastoma and other cancers and is associated with increased cell proliferation and tumor aggressiveness.
