Background

EGFRvIII is a constitutively active, ligand-independent mutant of epidermal growth factor receptor (EGFR), arising from the in-frame deletion of exons 2-7 in the EGFR gene. This deletion eliminates the extracellular ligand-binding domain, triggering spontaneous dimerization and persistent activation of oncogenic signaling pathways (e.g., RAS/RAF/MEK/ERK, PI3K/AKT/mTOR) that drive cell proliferation, survival and metastasis. Uniquely absent in normal tissues but abundant in multiple malignancies (e.g., glioblastoma, non-small cell lung cancer), EGFRvIII serves as a highly attractive target for cancer diagnosis and targeted therapy.
