- Mycobacterium tuberculosis HSP65 (Heat Shock Protein 65) is a chaperone protein of Mycobacterium tuberculosis, belonging to the highly conserved HSP60 family of heat shock proteins. Its primary function is to assist in the folding, refolding, and assembly of other bacterial proteins, especially under stress conditions (e.g., oxidative stress, heat, or nutrient deprivation) encountered within the host. This chaperone activity is critical for maintaining bacterial protein homeostasis and survival during infection. HSP65 also has strong immunogenicity, as it is recognized by the host's immune system, stimulating both B-cell and T-cell responses. It is used as a diagnostic marker for tuberculosis, as HSP65-specific antibodies are often detected in infected individuals. Additionally, it is a candidate antigen for tuberculosis vaccines, though its homology to human HSP60 requires careful evaluation to avoid cross-reactive immune responses.
