Recombinant Mouse Antibody clone BB128, which is specific to HIV-1 p24. BB128, recognizes the p24 loop sequence EAAEWDRVHP.

HIV-I viral lysate proteins were electrophoresed on a 12% SD-polyacryiamide gel and transferred to a membrane from which identical strips were prepared. Each strip was probed with 1 μg/ml of the mAb indicated, then incubated with alkaline phosphatase-conjugated goat anti-mouse IgG, and developed. Molecular weights (in kilodaltons) of protein markers run on the same gel are indicated with arrows.

A constant amount of fluorescein-labeled mAb was bound to excess antigen (HIV-I lysate) in the presence of serial 2-fold dilutions of unlabeled competing mAb. The fluorescein-labeled mAb is used at a concn corresponding to approximately half its saturation value in a PCFIA direct binding assay, while the highest concn of unlabeled mAb corresponds to its near-saturation value in a PCFIA direct binding assay. Values plotted are means of values from triplicate assays. Symbols for the unlabeled mAbs in all graphs are: ○ BI1279, ● BB128, + BB328, × BB223, □ BB344, ■ B11710, and ∆ BI1777. Fluorescein-labeled mAbs are (A) BI1279; (B) BB128; (C) BB223; (D) BB344; (E) BI1710; and (F) BI1777.

Primary antibodies used at 1 μg/ml were mAbs (A) B11279, (B) BB128, (C) BB328, and (D) BB223. Three other mAbs produced patterns similar to that seen with BB223: BB344, B11710, and B11777, although this final antibody was used at a concn of 10 μg/ml. Even with the higher concn of BI1777, binding of this mAb to the p24 fusion protein on western blot was quite weak. Although BI1777 clearly binds to p24005 and p24006 samples, its pattern of weak binding is less evident and may not be identical to that of mAb BB223 in (D).
