The anti-HSPB1 pH-dependent antibody (VS-0825-YC158) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human HSPB1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for HSPB1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from HSPB1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for HSPB1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human HSPB1, or Heat Shock Protein B1, is a small heat shock protein that is involved in protein folding and cellular stress response. It is often overexpressed in cancer cells, where it contributes to drug resistance and cell survival.
