The anti-IDO1 pH-dependent antibody (VS-0825-YC162) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IDO1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IDO1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IDO1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IDO1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IDO1, or Indoleamine 2,3-dioxygenase 1, is an enzyme that catabolizes the amino acid tryptophan. It is expressed in various immune cells and its activity can suppress the immune response, making it a target for cancer immunotherapy.
