The anti-IL19 pH-dependent antibody (VS-0825-YC180) targets human IL19 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL19 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL19 clearance. The antibody operates in a cyclical manner, binding IL19 in the extracellular fluid and releasing it in acidified endosomes. Human IL19, or Interleukin-19, is a cytokine of the IL-10 family that is involved in inflammation and has both pro- and anti-inflammatory properties. It is a key player in skin inflammation and is also linked to the progression of certain cancers.
