The anti-IL36RN pH-dependent antibody (VS-0825-YC199) targets human IL36RN in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL36RN acid-switched antibody was engineered by incorporating histidine residues into its variable regions. These histidine substitutions within its Complementarity-Determining Regions (CDRs) enable pH-responsive antigen binding, which leads to enhanced IL36RN clearance. The antibody operates in a cyclical manner, binding IL36RN in the extracellular fluid and releasing it in acidified endosomes. IL36RN (Interleukin-36 Receptor Antagonist) is a cytokine that acts as a natural inhibitor of IL-36 signaling, playing an important role in controlling skin inflammation and regulating the immune response, particularly in diseases like psoriasis.
