The anti-Integrin αvβ6 pH-dependent antibody (VS-0825-YC366) targets Integrin αvβ6 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin αvβ6 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin αvβ6 clearance. The antibody operates in a cyclical manner, binding Integrin αvβ6 in the extracellular fluid and releasing it in acidified endosomes. Integrin αvβ6 is a receptor for fibronectin and is a key activator of latent TGF-β. It is involved in epithelial cell migration and is a crucial player in the development of fibrosis and cancer.
