The anti-MMP12 pH-dependent antibody (VS-0825-YC253) targets both human and mouse MMP12 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-MMP12 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced MMP12 clearance. The antibody operates in a cyclical manner, binding MMP12 in the extracellular fluid and releasing it in acidified endosomes. Human MMP12 (Matrix Metalloproteinase 12), also known as Macrophage Elastase, is a protease that degrades components of the extracellular matrix. It is involved in tissue remodeling and inflammation and is implicated in diseases like emphysema and atherosclerosis.
