SmartAb™ Recombinant Anti-MMP13 pH-dependent Antibody (Clone 14D10)

The anti-MMP13 pH-dependent antibody (clone 14D10) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Matrix Metalloproteinase 13 (MMP13). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MMP13 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MMP13 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MMP13, also known as Collagenase 3, is a protease that degrades collagen. It is a key player in osteoarthritis and is often overexpressed in various cancers, promoting tumor invasion and metastasis.
