The anti-NGF pH-dependent antibody (VS-0825-YC266) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human NGF. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NGF under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NGF at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human NGF (Nerve Growth Factor) is a neurotrophic factor essential for the survival, growth, and differentiation of neurons. It is involved in pain signaling and is a therapeutic target for conditions such as chronic pain.
