The anti-TIGIT pH-dependent antibody (VS-0825-YC329) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TIGIT. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TIGIT under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TIGIT at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TIGIT (T cell immunoreceptor with Ig and ITIM domains) is a co-inhibitory receptor expressed on T-cells and natural killer (NK) cells. It functions as an immune checkpoint molecule that suppresses immune responses, making it a promising therapeutic target for cancer immunotherapy.
