The anti-TNFRSF13C pH-dependent antibody (VS-0825-YC336) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human and cynomolgus TNFRSF13C. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF13C under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF13C at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. TNFRSF13C, also known as BAFF-R, is the receptor for the cytokine BAFF. It is essential for the maturation and survival of B-cells, and its signaling is implicated in autoimmune diseases.
