The anti-TNFSF4 pH-dependent antibody (VS-0825-YC347) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD252. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFSF4 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, mAbs that showed a significant increase in dissociation from TNFSF4 at pH 6.0 were identified. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFSF4, also known as OX40L or CD252, is the ligand for the co-stimulatory receptor OX40. It is involved in T-cell activation and is a potential therapeutic target for cancer immunotherapy.
