Anti-UBE2A Antibody Prodrug, Protease Activated (AA2)

The Anti-UBE2A Antibody Prodrug, Protease Activated (AA2) is an antibody prodrug comprised of a recombinant human antibody targeting human UBE2A, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to UBE2A protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to UBE2A effectively. UBE2A is an E2 ubiquitin-conjugating enzyme that is an oncogene when fused to the TFG gene, forming the TFG-UBE2A fusion found in a subset of myeloid malignancies. This fusion protein promotes oncogenesis through aberrant ubiquitination.

  • The modification of proteins with ubiquitin is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, ubiquitin-conjugating enzymes, and ubiquitin-protein ligases. This gene encodes a member of the E2 ubiquitin-conjugating enzyme family. This enzyme is required for post-replicative DNA damage repair, and may play a role in transcriptional regulation. Mutations in this gene are associated with cognitive disability. Alternative splicing results in multiple transcript variants.
