ARG1: A Key Metabolic Checkpoint in Tumor Immunity

Arginase 1 (ARG1) is a cytosolic enzyme that catalyzes the hydrolysis of L-arginine to L-ornithine and urea. ARG1 is best known for its essential role in the liver urea cycle but, it is also a major player in tumor immunity. Myeloid-derived suppressor cells (MDSCs) overexpress ARG1 in the tumor microenvironment and, through local arginine depletion, essentially starve anti-tumor T cells of this essential amino acid, shutting them down. Inhibition of ARG1 function is, therefore, a promising strategy to restore T-cell function and make other immunotherapies work better.

Background

Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exist (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type I isoform encoded by this gene, is a cytosolic enzyme and expressed predominantly in the liver as a component of the urea cycle. Inherited deficiency of this enzyme results in argininemia, an autosomal recessive disorder characterized by hyperammonemia. Two transcript variants encoding different isoforms have been found for this gene.
