CASP9: The Apex of the Intrinsic Apoptosis Pathway

As an initiator cysteine protease, Caspase-9 (CASP9) is a central regulator of the intrinsic apoptosis pathway. In response to various cellular insults and DNA damage, pro-apoptotic members of the BCL-2 family trigger the release of the cytochrome c from the mitochondria into the cytosol. In the cytosol, cytochrome c binds to a scaffold protein called Apaf-1, which in turn leads to the assembly of Apaf-1 into a large, wheel-shaped apoptosome complex. The apoptosome then recruits and dimerizes pro-caspase-9, thereby activating it. Activated caspase-9 then cleaves and activates the downstream executioner caspases, mainly caspase-3, committing the cell to die.

Background

This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein can undergo autoproteolytic processing and activation by the apoptosome, a protein complex of cytochrome c and the apoptotic peptidase activating factor 1; this step is thought to be one of the earliest in the caspase activation cascade. This protein is thought to play a central role in apoptosis and to be a tumor suppressor. Alternative splicing results in multiple transcript variants. [provided by RefSeq, May 2013]

Caspase-9 and APAF1 bind to each other via their respective NH2-terminal CED-3 homologous domains in the presence of cytochrome C and ATP. Interacts (inactive form) with EFHD2. Interacts with HAX1. Interacts with BIRC2/c-IAP1, XIAP/BIRC4, BIRC5/survivin, BIRC6/bruce and BIRC7/livin.
