Encoded by the CBX5 gene, it is a non-histone chromosomal protein. CBX5: The Heterochromatin Protein 1 Alpha (HP1α)
The Chromobox Protein Homolog 5 (CBX5) gene encodes the Heterochromatin Protein 1 Alpha (HP1α) which is one of the basic non-histone chromosomal proteins. HP1α is a structural component of heterochromatin (dense and transcriptionally inactive region of the genome). It is an epigenetic “reader.” HP1α has a chromodomain that specifically binds to histone H3 when it is trimethylated at lysine 9 (H3K9me3), a classical epigenetic mark for silent chromatin. HP1α then recruits proteins that compact the chromatin and ensure that genes remain off. This is crucial to genome integrity and stable repression of genes, for example, transposable elements.

Background
This gene encodes a highly conserved nonhistone protein, which is a member of the heterochromatin protein family. The protein is enriched in the heterochromatin and associated with centromeres. The protein has a single N-terminal chromodomain which can bind to histone proteins via methylated lysine residues, and a C-terminal chromo shadow-domain (CSD) which is responsible for the homodimerization and interaction with a number of chromatin-associated nonhistone proteins. The encoded product is involved in the formation of functional kinetochore through interaction with essential kinetochore proteins. The gene has a pseudogene located on chromosome 3. Multiple alternatively spliced variants, encoding the same protein, have been identified.
