CD96, a member of the immunoglobulin superfamily, is a critical regulator of immune cell function. It interacts with nectin and nectin-like proteins and, in conjunction with TIGIT, acts as a co-inhibitory receptor for CD226. This regulatory mechanism suggests that CD96 and TIGIT might represent helpful targets for cancer immunotherapy. Recent studies have demonstrated CD96's significance in modulating the activity of NK cells and CD8+ T cells, highlighting its pivotal role in tumor immune surveillance. Background

The protein encoded by this gene belongs to the immunoglobulin superfamily. It is a type I membrane protein. Alternative splicing generates multiple transcript variants encoding distinct isoforms.
