SmartAb™ Recombinant Anti-CD96 pH-dependent Antibody (Clone 6A6)

The anti-CD96 pH-dependent antibody (clone 6A6) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to mouse CD96. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD96 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD96 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD96 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Mouse CD96 is an activating receptor expressed on T-cells and NK cells that promotes cytotoxicity. It is involved in both anti-tumor and anti-viral immune responses, making it a potential target for cancer immunotherapy.
