CDK2: The Key Kinase for G1/S Transition
CDK2 (cyclin dependent kinase 2) is a serine/threonine protein kinase that plays a key role in regulating the cell cycle. The kinase’s activity is tightly regulated through its association with cyclins. CDK2 primarily binds to two major cyclins: Cyclin E, which promotes the G1/S transition, and Cyclin A2, which drives S-phase progression and entry into mitosis. Once activated, CDK2 can phosphorylate the retinoblastoma protein (Rb) and other substrates to override cell cycle checkpoints, allowing the cell to enter DNA replication. This kinase was once considered an essential cancer target, but its inhibition is complicated by functional redundancy with CDK1. Nevertheless, CDK2’s importance in proliferation, and its frequent dysregulation in tumors, have kept it in the spotlight for research and therapeutic development.

Background
Cyclin-dependent kinase 2, also known as cell division protein kinase 2, or Cdk2, is an enzyme that in humans is encoded by the CDK2 gene. The protein encoded by this gene is a member of the cyclin-dependent kinase family of Ser/Thr protein kinases.
