PDE2A: The cGMP-Stimulated Cyclic Nucleotide Phosphodiesterase
Phosphodiesterase 2A (PDE2A) belongs to the phosphodiesterase superfamily of enzymes, which catabolize the intracellular second messengers cyclic AMP (cAMP) and cyclic GMP (cGMP). PDE2A is the first cGMP-regulated cyclic nucleotide PDE that was purified and its biochemical properties described. As a dual-specificity enzyme, PDE2A can degrade both cAMP and cGMP. The activity of PDE2A is allosterically regulated by the binding of cGMP to its GAF domains. This leads to an intriguing signaling crosstalk: a rise in cGMP (e.g. resulting from NO signaling) results in the activation of PDE2A, which then causes a transient degradation of cAMP, providing important control of heart rate, adrenal steroid production, and neuronal activity.
