PDE4B: A Promising Therapeutic Target in Inflammatory Neurological and Cancer Disorders
PDE4B is an enzyme that plays a pivotal role in regulating intracellular cyclic AMP (cAMP) levels, which are critical for numerous cellular functions including inflammatory responses, immune regulation, and neural signaling. Aberrant PDE4B activity has been linked to a variety of diseases, particularly those involving inflammation and the nervous system. Elevated PDE4B expression is associated with chronic inflammatory diseases such as asthma, chronic obstructive pulmonary disease (COPD), and rheumatoid arthritis, where it contributes to excessive immune activation and tissue damage. In the brain, altered PDE4B function affects neurotransmitter signaling and has been implicated in psychiatric disorders including schizophrenia, depression, and anxiety, influencing cognitive and mood regulation. Moreover, PDE4B has emerged as a factor in cancer biology, where its dysregulation may promote tumor growth and metastasis through effects on cell proliferation and survival pathways. Background
This gene is a member of the type IV, cyclic AMP (cAMP)-specific, cyclic nucleotide phosphodiesterase (PDE) family. The encoded protein regulates the cellular concentrations of cyclic nucleotides and thereby play a role in signal transduction. Altered activity of this protein has been associated with schizophrenia and bipolar affective disorder. Alternative splicing and the use of alternative promoters results in multiple transcript variants encoding different isoforms.
