anti-ASTL immunotoxin D-8 (Fab)-Sap (CAT#: AGTO-L044S)
This immunotoxin was achieved by conjugating/fusing the Anti-ASTL Fab to saporin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to kidney cancer and pancreatic cancer cells and enters cells by receptor-mediated endocytosis. Upon release of the conjugate from endosomes, the saporin toxin catalytically renders the large subunit ribosomes unable to bind the elongation factors and inhibits protein synthesis. It was designed for treatment of kidney cancer and pancreatic cancer.

- Saporin is a so-called ribosome inactivating protein (RIP), due to its N-glycosidase activity, from the seeds of Saponaria officinalis (common name: soapwort).Saporin depurinate the ribosomal RNA in a specific position; rendering the large subunit of ribosomes unable to bind the elongation factors. The consequent arrest of protein synthesis is responsible for the cytotoxicity.
