The anti-CALCA pH-dependent antibody (VS-0825-YC27) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CALCA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CALCA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CALCA at pH 6.0. This pH-sensitive antibody retains its binding affinity for CALCA on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CALCA is the gene that encodes the precursor for calcitonin and calcitonin gene-related peptide (CGRP), two important signaling molecules. Calcitonin regulates calcium and phosphate levels in the blood, while CGRP is a potent vasodilator and is implicated in the pathophysiology of migraines.
