The anti-CCL11 pH-dependent antibody (clone Bertilimumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CCL11. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CCL11 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CCL11 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CCL11 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CCL11, also known as Eotaxin-1, is a chemokine that acts as a potent chemoattractant for eosinophils. It plays a central role in allergic inflammation and is a key mediator in diseases such as asthma and allergic rhinitis.
