The anti-CCL5 pH-dependent antibody (VS-0825-YC33) targets human CCL5 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CCL5 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CCL5 clearance. The antibody operates in a cyclical manner, binding CCL5 in the extracellular fluid and releasing it in acidified endosomes. Human CCL5, also known as RANTES, is a chemokine produced by a variety of cells, including T-cells and platelets, that attracts T-cells, eosinophils, and basophils.
