The anti-CXCL1 pH-dependent antibody (VS-0825-YC89) targets human CXCL1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CXCL1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CXCL1 clearance. The antibody operates in a cyclical manner, binding CXCL1 in the extracellular fluid and releasing it in acidified endosomes. Human CXCL1, also known as growth-regulated oncogene-alpha (GRO-α), is a chemokine that primarily attracts neutrophils to sites of inflammation. It is a key mediator in inflammatory responses and is also involved in angiogenesis and tumor growth.
