The anti-CXCL12 eliminating antibody is an engineered pH-sensitive antibody against human and mouse CXCL12. CXCL12 regulates diverse cellular roles and is linked to HIV resistance. The antibody has a novel CXCL12-sweeping activity in vivo, which selectively eliminatesthe soluble CXCL12 from plasma. Compared to conventional antibody, the mAb exerted in vivo efficacy by simultaneously engineering both pH-dependent antigen binding, to mimic the pH-dependent receptor-ligand binding of such endocytic receptors, and increased FcRn binding affinity at neutral pH, to mimic the cell-bound form of the receptors.
