The anti-CXCL12 pH-dependent antibody (clone 30D8) targets both human and mouse CXCL12 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CXCL12 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CXCL12 clearance. The antibody operates in a cyclical manner, binding CXCL12 in the extracellular fluid and releasing it in acidified endosomes. CXCL12, or stromal cell-derived factor-1 (SDF-1), is a chemokine that is essential for the homing of hematopoietic stem cells to the bone marrow and for the migration of various immune cells. It is also involved in organogenesis, angiogenesis, and cancer metastasis.
