- Treponema pallidum p47 is a core outer membrane protein of Treponema pallidum, playing dual roles in bacterial pathogenesis and host immune recognition. As an adhesion protein, it enables Treponema pallidum to colonize host mucosal surfaces, a necessary first step in syphilis transmission and infection. As an immunogenic protein, it elicits a robust antibody response that is central to syphilis testing-many commercial syphilis assays include p47 as a target to detect infection. The protein's conservation across Treponema pallidum strains ensures that these tests work for all clinical isolates, reducing false negatives. In vaccine research, p47 is valued for its surface exposure: antibodies generated against p47 can directly interact with the bacterium, blocking adhesion and neutralizing infectivity. While a syphilis vaccine is not yet available, p47 remains a leading candidate due to its role in key infection processes and strong immunogenicity.
