SmartAb™ Recombinant Anti-F9 pH-dependent Antibody (Clone 11B4-C3)

The anti-F9 pH-dependent antibody (clone 11B4-C3) targets human F9 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-F9 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced F9 clearance. The antibody operates in a cyclical manner, binding F9 in the extracellular fluid and releasing it in acidified endosomes. Human F9, or Coagulation Factor IX, is a vitamin K-dependent serine protease of the intrinsic coagulation pathway. Mutations in its gene cause hemophilia B, a bleeding disorder characterized by a deficiency in this clotting factor.
