The anti-FLT4 pH-dependent antibody (VS-0825-YC137) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human FLT4. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for FLT4 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from FLT4 at pH 6.0. This pH-sensitive antibody retains its binding affinity for FLT4 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human FLT4, also known as VEGFR3, is a receptor tyrosine kinase that is essential for lymphangiogenesis, the formation of lymphatic vessels. It is a key regulator of lymphatic vessel development and is also involved in the metastasis of certain cancers.
