The Anti-HGF Antibody Prodrug, Protease Activated (VS-1025-YC52) is an antibody prodrug comprised of a recombinant mouse antibody targeting human HGF, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to HGF protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to HGF effectively. HGF (Hepatocyte Growth Factor) is the ligand for the MET receptor and acts as an important factor in the tumor microenvironment. HGF secreted by stromal cells activates MET signaling on tumor cells, leading to enhanced cell motility, invasion, angiogenesis, and the metastatic spread of the tumor. Its high concentration in the tumor microenvironment is often associated with poor clinical outcomes.

- This gene encodes a protein that binds to the hepatocyte growth factor receptor to regulate cell growth, cell motility and morphogenesis in numerous cell and tissue types. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate alpha and beta chains, which form the mature heterodimer. This protein is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. This protein also plays a role in angiogenesis, tumorogenesis, and tissue regeneration. Although the encoded protein is a member of the peptidase S1 family of serine proteases, it lacks peptidase activity. Mutations in this gene are associated with nonsyndromic hearing loss.
