The Anti-HIF1A Antibody Prodrug, Protease Activated (6A9) is an antibody prodrug comprised of a recombinant mouse antibody targeting human HIF1A, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to HIF1A protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to HIF1A effectively. HIF1A (Hypoxia-Inducible Factor 1 Alpha) is a master transcription factor that drives the adaptation of tumors to low oxygen (hypoxic) conditions. Its stabilization and activity promote angiogenesis (via VEGFA) and metabolic reprogramming, which are essential for the survival and growth of solid tumors.

- Introduction
- This gene encodes the alpha subunit of transcription factor hypoxia-inducible factor-1 (HIF-1), which is a heterodimer composed of an alpha and a beta subunit. HIF-1 functions as a master regulator of cellular and systemic homeostatic response to hypoxia by activating transcription of many genes, including those involved in energy metabolism, angiogenesis, apoptosis, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia. HIF-1 thus plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease. Alternatively spliced transcript variants encoding different isoforms have been identified for this gene.
