The anti-HIF1A pH-dependent antibody (clone 1H1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human HIF-1alpha. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for HIF-1alpha under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from HIF-1alpha at pH 6.0. This pH-sensitive antibody retains its binding affinity for HIF-1alpha on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human HIF-1alpha, or Hypoxia-Inducible Factor-1 alpha, is a transcription factor that is a master regulator of the cellular response to low oxygen levels (hypoxia). It regulates the expression of genes involved in angiogenesis, metabolism, and cell survival.
