The anti-IGF1 pH-dependent antibody (clone MuEM164) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IGF1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IGF1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IGF1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IGF1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IGF1, or Insulin-like Growth Factor 1, is a hormone with structural similarity to insulin that is involved in cell growth and proliferation. It plays a key role in childhood growth and has anabolic effects in adults.
