The anti-IL17RA pH-dependent antibody (VS-0825-YC178) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL17RA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL17RA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL17RA at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL17RA on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL17RA, or Interleukin-17 Receptor A, is a transmembrane receptor that binds to IL-17 cytokines. It is a key component of the IL-17 receptor complex and is essential for IL-17 signaling, playing a critical role in inflammation and autoimmunity.
