The anti-IL1A pH-dependent antibody (VS-0825-YC181) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL1A. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL1A under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL1A at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL1A on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL1A, or Interleukin-1 alpha, is a pro-inflammatory cytokine that is a key mediator of the inflammatory response. It is produced by a variety of cells and plays a critical role in host defense against infections and in sterile inflammation.
