The anti-IL31RA pH-dependent antibody (VS-0825-YC195) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL31RA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL31RA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL31RA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells, while having negligible binding to IL31RA expressed on normal cells under physiological pH. Human IL-31RA, or Interleukin-31 Receptor A, is a transmembrane receptor that binds to IL-31. It is a key player in skin inflammation and is a therapeutic target for inflammatory skin diseases.
