The anti-Integrin α5β1 pH-dependent antibody (VS-0825-YC226) targets human Integrin α5β1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin α5β1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin α5β1 clearance. The antibody operates in a cyclical manner, binding Integrin α5β1 in the extracellular fluid and releasing it in acidified endosomes. Integrin α5β1 is a cell surface receptor for fibronectin. It is involved in cell adhesion, migration, and survival, playing a key role in angiogenesis and being overexpressed in various cancers.
