The anti-Integrin α5β1 pH-dependent antibody (VS-0825-YC364) targets human Integrin α5β1 in a pH-dependent manner that affinity is kept high at neutral pH but becomes weaker when pH is lowered, such as in the acidic environment of the endosomes. The anti-Integrin α5β1 acid-switched antibody was engineered by incorporating histidine residues into their variable regions. The introduction of histidine substitutions into its CDRs allowed pH-responsive antigen-binding leading to enhanced Integrin α5β1 clearance. The antibody works in cycles of Integrin α5β1 binding in the extracellular fluid and Integrin α5β1 releasing in acidified endosomes. Human Integrin α5β1, or Integrin Integrin α5β1, is a cell adhesion receptor that is involved in the binding of cells to fibronectin. It is a key player in cell migration and is involved in various processes, including wound healing and angiogenesis.
